LEVOBUPIVACAINE A long acting local anaesthetic, with less cardiac and neurotoxicity

چکیده
Stereoisomerism describes those compounds which have the same molecular formula and chemical structure, but the atoms are orientated in a different direction. There are two isomers, each a mirror image of the other, called enantiomers. They are also called optical isomers because they rotate the plane of polarised light either to the right referred to as +, dextro, d or D isomer, or to the left referred to as -, laevo (levo), l or L isomer. More recently this classification has been replaced by the R/Snotation, which describes the arrangement of the molecules around the chiral centre (R is for rectus the Latin for right, and S for sinister, left). The R enantiomer rotates light to the right and the S enantiomer to the left. As with other isomers, they can have different properties.

TOXICOLOGICAL STUDIES ON CERTAIN AMINOALKYL ETHERS AS POTENTIAL LONG-ACTING LOCAL ANAESTHETICS
The acute intravenous toxicities of a selection of three potential long-acting local anaesthetics belonging to the series of2- phenoxyethyldialkylamine were determined in mice. Two of the compounds were tertiary amines and the third a quaternary ammonium derivative. The I.V. median lethal dose, (LDso) of one of the tertiary compounds, 21A [N, N- diethyl-2- (2,6- diisopropyl) ethyl amine hyd...
متن کاملMinimum effective local anaesthetic dose of isobaric levobupivacaine and ropivacaine administered via a spinal catheter for hip replacement surgery.
BACKGROUND Continuous spinal anaesthesia with spinal catheters allows incremental dosing of local anaesthetic and, consequently, less haemodynamic changes. However, little is known about the required doses. Therefore, we designed a study to assess the minimum effective local anaesthetic dose (MLAD) of levobupivacaine and ropivacaine in this context. METHODS Forty-one patients undergoing hip r...

AIMS The Authors examine the employement of a new anaesthetic agent, levobupivacaine 0.50% (S - enantiomer of racemic mixture of bupivacaine), for peribulbar anaesthesia in a randomized double blind study vs. racemic bupivacaine 0.50% alone or in association with hyaluronidase 10 IU x ml(-1). MATERIALS AND METHODS 120 patients were divided into four groups of 30 each: group L (levobupivacaine...
متن کاملLocal anaesthetic sensitivities of cloned HERG channels from human heart: comparison with HERG/MiRP1 and HERG/MiRP1 T8A.
BACKGROUND Myocardial potassium channels are complexes formed by different subunits. The subunit composition may influence the cardiotoxic action of local anaesthetics. The effects of amide local anaesthetics on HERG channels co-expressed with the putative subunit MiRP1 have not been established. It is also unclear if the common polymorphism MiRP1(T8A) that predisposes individuals to drug-induc...
متن کاملA NEW BIOLOGICAL SCREENING SYSTEM FOR LOCAL ANAESTHETICS BY INHIBITION MOBILITY OF TETRAHYMENA PYRIFORMIS
An alternative in vitro approach to drug screening has been the use of human cell cultures for antiviral agents and microbial cell cultures for the assessment of the carcinogenic potential of selected compounds. A number of protozoan species have been also used as drug screens for anti-protozoal agents. The ciliated protozoan Tetrahymena pyriformis species has been widely utilised as a dru...
